Symptomatology and Microecology of Pseudomonas tolaasii Infection in Button Mushroom

چکیده
Brown blotch disease caused by Pseudomonas tolaasii represents a major biotic stress affecting the physiological integrity of cultivated mushrooms. This study aimed to evaluate the pathogenicity of P. tolaasii on Agaricus bisporus and to investigate infection associated ultrastructural damage using scanning electron microscopy (FE-SEM). Pathogenicity assays were performed on fresh mushroom caps using the cap-cut method, followed by symptom observation and bacterial re-isolation. Infected caps developed typical brown blotch lesions within 48 h, whereas control tissues remained symptomless. FE-SEM analysis revealed dense bacterial attachment to hyphal surfaces, accompanied by membrane degradation and pore like structural damage, which were absent in control samples. These findings indicate that P. tolaasii induces disease through hyphal colonization and membrane disruption, leading to physiological failure in mushroom tissues.
